I have a standard Mexi Strat and an ibanez TS-9. I am trying to get the good bluesey setting. Like the "Couldn't Stand The Weather" sound. What settings do you have for your Tube Screamer?

What kind of amp are you feeding this signal into?? Those settings will constitute part of your final tone.

Use the neck pickup. tweek the TS-9 until you get a creamy sustain, and then tweak your amp, heavy in the mid-range. Any overdrive from the amp should be just for an edge because your getting it from the TS-9 acting like a midboost. You might find it more to your liking keeping the amp clean.

There's a tendency these days to overfuzz and distort the hell out of
everthing until it sounds like mud.

well the most important thing to get good sound out of it is to play it trough a tube amp.
my favourite setting:
Drive:10,30 O'clock
Tone:11
Level:different from time to time.....

You've those presets in the Line 6. Just mix'n match until you find your lost chord. You might want to start with a Bassman or Twin preset if you have it. .

A Spider and a Crate?

In my experience Line 6 overcook everything hugely on the FX side. (That probably goes for all manufacturers of digital sims units.) You might want to turn everything down from the factory presets a little: it'll sound a bit thin to your ear at first, but give it a while and you'll discover that is where the subtlety of a fine blues sound comes from.

Nearly always, the pros you admire use a lot less of, well, everything really, than you might imagine.

all you need is the tube screamer combinded with just a bit of gain from the amp for filler and youll get the blues sound, some people use 2 tube screamers, personally i use a pinch of boost from my amp and a tube screamer. It sounds amazing

I don't think you're going to get there with a tube screamer. SRV didn't use them all of the time. For the rhythm parts, it sounds like it's just his amp, and maybe he kicks on the TS for the solo. The amp really is the most important part of his tone...old Fender 40 watt tube amps.

Gotta go with Doc on this when the TS 9 has been a main stay of mine for years and I find that it sounds better with the tone @ 3 oclaock and the drive a little above 2 oclock. People do tend turn the drive all the way up and you end up with a distortion wash instead of the nice break up at lower settings

One thing I notice when running my stratocasters through a TS-808 Tube screamer is it always sounded so brittle and dry. Then I got Holy Grail Plus reverb pedal and it made a world of difference.

If you really want to get closer to the SRV sound, you have to remember that he always tuned down and used heavier gauge strings(.011 I think) to get a nice fat tone without a ton of distortion. Overdrive would be more the sound from his old Fender 40 watt tube amps.
Check out some of the old Austin City Limits performances to get an idea of what is goin' on!!!

I'm running through a Twin Reverb RI and have my TS-808 (slightly different tone and it's modified by Keeley) set at Drive 3:00, Level 9:00, Tone 11:00.
