I believe the tone you're referring to is from a compressor.

Some compressors (such as the dyna comp) add a bit of their own tone to the guitar.

You can achieve a similar result with an on-board active mid boost circuit such as Fender's MDX and EMG's SPC.

Clapton refers the MDX midrange circuit on his guitars as a "compressor", which is a bit confusing because a real compressor has nothing to do with an internal mid boost preamp despite their similarities function-wise.

Agree all the way to this point. That tone is definitely not pickup dependent. You need to know his signal and effects chain and the EQ for that song. Rotza-ruck :!: The closet I ever came to that kind of luck was sitting literally three feet from Les Paul's feet, at the Iridium in NYC, about 6 months before he died. With his floor board right in front of my nose, I was too busy focused on the man to pay much attention to the settings on his stomp boxes. Not to mention the fact that his guitar had been created by the Borg Collective. Resistance was futile.

Sounds compressed to me though, but I'm no pro.

He is a great player.
That's pretty much the sound of a good bright neck pickup on a Strat into a slightly dirty tube amp. (with wah in some parts).
A lot of people EQ their amp for a fat bridge sound so the neck sounds muddy.
But if you set for you neck pickup it can have this sweet tone (skill aside)
