The 'in between' positions on a Strat (2 and 4, the neck/mid and mid/bridge combined) are what mark out the biggest difference for me... although, traditionally a Tele will have very different resonant qualities due to not having a large amount of wood carved out for a trem unit.

Horses for courses... play both and have a look. All I remember about the two Telecasters I owned is that one had a lovely smooth neck pickup (Lite Ash Seymour Duncan) that suited almost everything, and that humbuckers in a Tele may not be the answer to 'beef it up' (American Deluxe FMT HH Tele).

I remember my Strats a lot more, and find the tones more memorable- to my ears, anyway. Which explains why I've had four times more Strats than Teles.
