My 54 japanese reissue is made out of basswood (I think). Can anyone tell the difference in tone between the different wood types?

I think pickups, intonation, string height, neck relief, pickup height... etc. play a much larger role in how a guitar sounds. I used to think the fretboard played no role but I can see now that it does. And I had a twin pivot bridge with those high mass saddles - which made my guitar sound :?.

Lot's of different things factor in that determine how a guitar feels and sounds - most of which will be lost on most people.

Wood type and construction certainly makes a difference in acoustic guitars but in electrics? I'm not sure.

Don't change a knob setting an go from one amp to another you get a different sound!

That is so true.

And ... if you turn the knobs on favorite amp, the tone of your Stratocaster's wood also changes. Try moving the pickup selector switch ... that REALLY changes the tone.
