I just got a Classic Vibe 50s Stratocaster in the 2-tone sunburst color, but I'm thinking of trying to get a black pickguard so it'll look a little more like this.

I've looked on Squier's website and on Amazon, and it looks like it needs an 8-hole type of pickguard in 1-ply, but they only have 8-hole 3-ply on Amazon, will that work?

Any other suggestions for mods?

Yup it should work. The screw holes are usually pretty close so long as it's the same number. As for the ply's, that's just a thickness/decorative feature as the middle ply is usually the opposite color of the black or white pickguard color.

If you're after a classic '50s vibe (see what I did there?) you have to go for single ply. To my eyes there is quite a difference in appearance between single and triple ply; and if you anchor your fingers on the scratchplate, the difference in height is actually quite noticeable IMHO.

You will still probably need to drill a new hole, when I put an aged "fender" branded (vintage 54 I think) 8 hole single ply reissue guard on my 50's Vibe, the screw closest to the pups switch is in a different location

Note the screw by the switch, stock its almost inline left to right with the switch screw, on the replacement its a pronounced diagaonal.
