Is the middle pickup on new American Standard Strats reverse wound, reverse polarity? If not, do I reverse the wiring to achieve the hum cancelling effect.

Yes the new Am Std middle pickup are still RW/RP just like their forefathers the Am Series and old Am Std. You're good to go.
