Well to start...

The Pick-Ups, the Bridge, the tuners, the ply (3,5,7,ect) in the body, the craftsmanship, the types of wood used... They are a different animal.

I have owned 2 MIM stratocasters. The first a Standard. Never really did it for me, the tuners wiggled, the nut moved, the pick-ups were weak. But it was a great learner. My current MIM is a 70's Re-production. Not a problem with it. The neck is smooth, the pick-ups get it on and the nut, stays in place.
My only issue are the volume and tone pots. They have a little play, but not bad at all...

I have a American "Fat-Strat" it's a 96, kinda beat up. The bridge and pick-ups really let you know it is very different from the MIM's. The string tree is different and the action is like nothing else. I got it used about 5 years ago and like I said, it's kinda beat up, but when you hear it and play it, ascetics go right out the window, this thing is "Sweet" and soooo effort less to play.

I used to really knock the MIM line, however, my 70's Model has changed my mind. It may not have the same bridge, pick-ups and tuners as my American, but it's got the Mojo... and that's what matters to me.

I used to have a MIM ('98 model I think). I didn't like the MIM. I kept having to adjust the action, never stayed in tune, and like the previous poster said, the pick ups sound different.

I have two Americans now and there is a huge difference. If you stick wtih playing the guitar and start gigging, I highly recommend getting an American--they're well worth it.

For what it's worth, the Deluxe series of MIM guitars is very good. I have a Deluxe Powerhouse Strat with the 12db boost that I absolutely love. It gets more time than my American series Strat (but not more than my Hwy1).

One is made in America by Mexicans..the other is made in Mexico by Mexicans. Basically what I can tell, it's the sum of the parts, tuners, bridges, pickups are lesser quality. It may hit or miss as to whether you an get a great MIM axe.

I have played Mexicans, Japanese, and USA, and the USA do just feel like a more solid guitar. That being said, I think the Mexicans and Japanese are still very good guitars. I think they are great value for their money. However, the reason I would purchase a MIA guitar, is that they hold their value (just look on eBay). In 20 years, your MIA Strat will be worth much more than your MIM or MIJ. But if you can't afford an MIA, the MIMs and MIJs are fine guitars!
