What are the major differences between the trem bridges of the American Standard and the American Deluxe Strats? Is one significantly better than the other? How would they compare to a Wilkinson?

Is a 2-point fulcrum more stable than a 6, and if so, why is that?

A truckload of great players have done fine with six-screw trems. Nothing wrong with them.

However, two-point trems tend to give a smoother motion to the vibrato which can feel and sound nice. And it is a little easier to adjust the overall height of the bridge. Also, the fulcrum blade on the six-screw type has been known to catch on those screws preventing the trem from returning to pitch correctly. It is addressable, but modern two-points arguably function a tad better.

Between the American Standard and Deluxe: the former currently has reverted to bent steel saddles (supposedly better for twang), the Deluxe has block saddles (thought better for sustain). And the Deluxe has a pop-in whammy arm, the Standard a screw-in. The Deluxe has slightly stronger strings, which makes no difference whatsoever once the guitar is setup. I don't know if they have different blocks.

Yeah, I put the Deluxe's block saddles on mim MIM Std. Very nice.

You did the Callaham block as well, didn't you, 01GT? Did that work out well for you?

Not the Callaham block, though. I went with the 06+ Fender OEM block (the 05s and before MIMs had a much smaller block). It was hard to say how much improvement because even with the smaller block, the sustain was excellent. Shortly after that mod, I did the deluxe saddles and Tex Mex pups. With all that, sustain and staying in tune are both excellent. Here is a pic of the saddles:
