Is a person suppose to use a reverse wound single coil pickup in a HSH configuration or are you suppose to use a regular single coil?

Depends on how you have it wired.
If you have it wired so the 2 and 4 positions split the bridge or neck positions and bring in the middle, then you would want the middle pickup RWRP relative to the active coil on the neck and bridge.
If you have no coil splitting and it's just a straight B/BM/M/MN/N, then it won't matter.
