Is it possible to use (2)250k pots for the tone and a 500k for the volume?
If not can someone explain why?

You could, but the 500k volume pot will brighten the tone, you may not care for it.

Yep, what he said.

CS'69s are very bright even with a 250K volume pot. With a 500K volume they'll border on being brittle/harsh sounding.

CS'69s are also fairly low output. If you're considering a 500K volume to get a little more output, yes that would give a little boost. But a clean boost pedal would give you way more output, without changing the tone.

If you're considering this simply because those are the pots you already have on hand, I'd lean toward using the 500K as the neck pickup tone control.

For some reason, G&L decided to go with 1m pots on the Climax, which was most often configured as a H-H model (pickups comparable to the Duncan Distortion), but they kept the same post configuration on the H-S-S model... partly due to the fact that the single coils were generic Asian OTC parts, and mainly due to the 1m pots, the single coil positions in that guitar were basically useless.
I really think G&L assumed that buyers of that configuration would be swapping in stacked or "rails" style humbuckers.

I'll bring a second opinion, and suggest you try it.
The possible advantage of a 500K vol pot is that turning the vol down doesn't roll off the highs as much as on a 250K - and only you know if that suits you or not.

There was a period in the '70s when Fender was putting 1 meg pots in Teles, to super-emphasize their brightness. And there were and are people who like that set up.

Since the original question was whether it's "possible" to use a 500K volume with 250K tones, yes it's possible.

We were telling you why most sources recommend 250K pots for Strat single-coils, and why most people are happiest with 250K pots.

But Jmattis is right. It can't hurt to try, and you might like it.
------------------
Context in the original question (or in a follow-up response) would have been helpful.

Why are you considering mixed pots? Is your Strat through your amp/pedals too dark sounding?

Or is it a purely theoretical question?

I'm still debating which way to go on my Partscaster.
I'm using current generation "hot" noiseless pickups, and originally intended to use the Greasebucket circuit, but someone mentioned that Fender uses 1m pots with the Noiseless... but in perusing the Strat service drawings, I'm seeing a lot of 250k on the non-active designs.
1m scares me. I'd like to come close to my '78, which I had replaced the stock pickups with SDS-1's.

hello, i am new here - too poor for fender -not going beyond 250k for single coils now, personally.

i have a S-S 7.5k 500k vol / tone (both), and it is very bright and harsh to my ears (even on middle / both "humbucking").. going to have to re-wire.

500k+ if your humbuckers aren't growling imho.

...can always mess about with tone capacitance but! i "can't figure out which way around to solder the capacitor" anyway... (most of the time).
