Which end of the cable is plugged into what, while you're holding the other end?

Is your external 12" speaker a passive monitor, or active - that is, does it contain its own power amp?

Have you bypassed/disconnected the speaker in your amp, or is the jack socket you've added in parallel or series with the existing speaker?

Sounds like a bad connection causing interference. Similar perhaps to if you plug a lead into an amp's input, then touch the plug at the other end of the cable (the end that would go into your guitar) - but louder because it's the amplified signal rather than the input signal.
