Hi Fender,
I have just connected the headphone outputs of my Mustang III v1 to my RME 400 inputs (channels 5+6), and sounds great. My question is though, the RME has settings for LO Gain, +4, or -10 for the inputs channels 5-8. Which setting is the correct one to match the headphone output on the mustang? I have it set on +4 at the moment, sounds perfectly fine to me, but just want to know if this an issue, or does it not matter?
Thanks,
Gary.

The "-10" sounds like a reference to "-10dBV" which is the line-level for "consumer equipment" and corresponds to 0.447 V peak (or 0.316 Vrms for a sine wave).

The "+4" sounds like a reference to "+4dBU" which is the line-level for "professional equipment" and corresponds to 1.737 V peak (or 1.228 Vrms for a sine wave).

So, one thing to try is to put an AC voltmeter across the amp's headphone output, and see what the voltage is (make sure you know whether your meter is showing RMS, peak, or even peak-to-peak which is double the basic peak number) while playing the guitar with the amp cranked to your desired maximum volume. If the answer is less than about 0.5V peak, use the "-10" setting. If the voltage is higher, use the "+4" setting.

I have no clue what the "LO gain" setting on your equipment does.

Or the other, simple way of doing this is just try each of the three settings on your RME400 to see which one sounds best/right: that is, not too quiet, but not so loud as to cause distortion/clipping. In each case, start with the amp volume low and slowly turn it up to your desired maximum volume, stopping if you start to hear distortion, to avoid risking any damage from overloading any of the equipment.

There's also the question of impedance-matching the amp's headphone output with the RME400's input, although this is less critical for line-level audio signals (where voltage is important rather than current and the wavelength is very long so unlikely to interact with cable length) than some people think. The key thing for best transmission (without any loss or distortion) of line-level audio signals, is that the RME400's input impedance should be much higher than the amp headphone socket's output impedance, which it almost certainly is, and keep the interconnect cables as short as possible.

Hey Scott, thanks for replying. I don't have a voltmeter (yet). Yeah, I have pretty much just gone for doing it the simple way :).
I have been trying the different settings and experimenting with levels on both the mustang and the RME. I have settled on the LO gain setting on the RME for now, as it seems 1) the safest, and, 2) to my ears anyway, it seemed to provide the cleanest sound albeit also the quietest level.
I'll stick with this for now, I'm happy with the sound I'm getting.
Interestingly, using the cab emulation via the headphone out into my computer via the RME is giving me the closest experience of playing through a real amp and speaker I have had with any modeling amp, and I have Amplitude 3 including the Fender pack, scuffham s-gear (which is very good btw), guitar rig 3, as well as Ampire which is included in Studio One 2.

Not using Fuse though.
Thanks,
Gary.
