Hi. I have just bought a Mustang Floor that I am running through a Roland Cube 40. What settings should I use on the Cube 40 to get the best out of the Mustang Floor?

It depends what settings (amp models and effects) you want to use on your M-Floor.

If you want to use the M-Floor's amp modelling, then you should set your Cube to a neutral sound. I don't know exactly what setting that is on the Cube, you may have to ask that on a Cube forum.

If you want to use the M-Floor's effects but not its amp modelling, then make sure the amp modelling is turned off on the M-Floor, and select appropriate models on the Cube.

Also make sure you haven't set up conflicting effects on the two devices, and that you haven't done anything silly with the signal chain ordering (eg putting a tremelo before an overdrive effect almost certainly won't give the overall effect you were intending). If you're not sure about that, there are lots of articles on the 'net about the "correct" order of different effects in a signal chain, and they mostly agree; my advice is to stick with such a recommended order until you know enough to experiment.

Finally, make sure you've set the M-Floor's output to match the way the Cube is setup; for example the M-Floor has different output 'voicings' depending whether you're going into a clean amp, a PA, etc. There's no absolute right answer here; just experiment with the options until you find one you like.

Finally finally, note the 'cab emulation' of the M-Floor is different to its setting for what you're plugging it into. Make sure you understand the difference so you can set them both appropriately.

I will say that you want make full use of the Mustang and use the cube only as power amp.

Basically set the tones at 12' o clock but if you want to mix better with other instruments or backing track just roll back a bit the basses and boost a bit the mids.

Thanks to Scott-UK and Mordor 74.
So basically JC Clean with no effects, and Treble Mid and Bass at 5?
Now to be a bit specific. I play a lot of different styles, and some of the names of the presets are pretty obvious - live and dangerous and cold sweat for Thin Lizzy, Johnny Marr for the Smiths, Mick the Hoople being Mick Ronson I presume, etc. But is there any definitive list of what the effects are based on?
I would like to know, for example, what is the best setting for
Zeppelin - Stairway to Heaven
Nirvana - Come As You Are
Guns n Roses - Knockin On Heaven's Door
Muse - Starlight
Leonard Cohen - Suzanne
Melanie Fiona - Monday morning
Amy McDonald - This is the life
Bruce Springsteen - The River
The Eagles - Hotel California
The Pogues - Dirty Old Town
Quite a spread, I think you will agree.
Bear in mind that the whole modelling thing is new to me!
I have no idea what cab emulation is.

No, unfortunately not. As you surmise, some of the factory presets are quite clearly intended to emulate well-known guitar/amp/guitarist sounds. One assumes the Fender engineers researched this to obtain good accuracy. What exactly they have done is not something they publish though - which is a shame as the info would be interesting (I suspect there are potential legal/copyright implications that prevent them).

Unless someone has already worked this out for themselves and is willing to post details of the presets they created, people on this forum probably won't know the answers to this any more than you. What I find works best is to research (lots of info on the net!) what actual guitars, amps, effects, etc, the musicians used for the original recording. Then dial-in comparable settings on the Mustang and tweak until it sounds good to you.

It's part of the amp modelling in the Mustangs (and also in other similar types of unit). Amplifiers have all sorts of different speaker cabinets, that all sound different: 1x12, 2x12, 4x12, 1x10, combo, separate head/cab, different speakers, etc. "Cab emulation" is an attempt to mimic the different sounds of those speaker cabinets.

For example, the "British 60s" amp model is based on a VOX 30, so has a "2x12" cab emulation as standard. But you can vary the sound by selecting different cabinets - personally I use the "British 60s" with the "57DLX" (Fender '57 Deluxe) cab, just because I like the sound that gives.
