Which Mustang model? I don't know how the M 1/2 do it, but on the M 3/4/5 it's a chromatic tuner: it tells you what pitch you're tuning to.

So just tune to D# - G# - C# - F# - A# - D# in the display (either on the amp or on the footswitch if you have the MS-4) and you're done. FUSE displays the same.

Although personally, I find it easier to use a separate tuner that I keep clipped to the headstock - eg search the web for "snark tuner"

The Mustang I and II are chromatic tuners as well. The Preset LEDs all have letters next to them: A, B, C, D, E, F, and G. Then the last preset has #, indicating that you're tuning to A# or B#, etc. Then the Red-Green-Red LEDs above the MOD dial show you how in-tune you are. Green by itself means that you're in tune at the note indicated by the preset LEDs. If you're a bit flat, it will show the left Red LED. As you get closer, the green will light up as well, then the red LED will go off. If you're too sharp, the RIGHT LED will be on. If you keep going, you'll switch to being flat on the next half-note up.

Just play around with it to learn how it works.
