On my M3VII, I use a Deluxe Reverb patch. On a standard Deluxe Reverb there is not a gain or a master, just a volume. But on the Mustang it has all 3 of these knobs. What do these knobs in the Mustang do? I know the master is the overall volume but what does the Gain and Volume knobs represent. Gain is usually preamp adjustment but the Volume is what? Does this knob effect the tone or is it just the line out volume?

VOLUME: changes the volume of only the currently selected preset

MASTER: changes the volume of ALL presets on the amp

So for example, let's say you set up 2 presets. You turn on the amp and load one of the presets. Now you move the MASTER to a comfortable position. But when you change to the second preset, it's too loud. Instead of adjusting the MASTER again, change the VOLUME of that preset to match the other preset.

The Mustang Amps' physical Master Volume knob (7th knob) controls the Mustang's final power amp stage volume level -- not the Preset's selected modeled amp's Power amp stage.

Depending on the circuitry of the modeled amp (profile) selected for the current Preset in use in your Mustang III v2, the physical Volume knob may or may not have much of an affect on the overall modeled amp's tone. It all depends on how the original inspired by amp's circuitry was designed.

Also note that a few of the amps, in their Advanced Amp settings (Press Amp Button 2x) also have a virtual Master Volume parameter.

With as many subtle variations amongst numerous similar Fender Amps and different channels in some of those amps, etc., plus factoring in the non-Fender amps, the best approach is often to let your ears guide you. Sweep through the Gain, Volume, virtual Master (advanced amp) Volume, if offered, to find the sweet spot(s) for your particular guitar(s), pickups, external pedals, if any, to suit your preferences.
