I'll give it a try -

I have two 250-210's and I set them identically as a starting point. The Semi-Parametric Equalizer has Low-Mid and High-Mid frequency select controls as well as a level control for each range. Each frequency control sets the center of optimum frequency response for it's range and the level control centers the output for the range in the amp so the mid-high and mid-low notes are at the same audio level as the deepest lows. Flat is a 12 o'clock setting for each control. Basically, those EQ controls are a fine tuning adjustment to correct for any frequency response issues that your pickups may have in being either too loud or not loud enough in the mid range. However, the mid range EQ controls can also be used to enhance the 'bright' jazz or rock style mids or reduce the mids for traditional country and similar styles and produce a deeper, rich bass..

You didn't mention the type of bass guitar you are using, so a good starting point would be to set your guitar tone control at mid range, set the low-mid EQ control to 700 hz and the mid-high EQ control to 2K . Then, set each of the EQ level controls located below the frequency controls to the detent at 12 o'clock position. Give it a try that way. If your ears tell you that the audio level for the mids is the same as for the deep lows, leave the settings there. You can also rotate the Treble control, located just to the right of the EQ controls up or down range to correct for high note roll off or to add high frequency response above 4 khz. For instance, a high 'D' note is roughly 4968 hz. If it is at a lower output level than your mids, rotate the treble control out of the 12 o'clock detent toward the +15 side. It takes some experimentation to get things right.

Also, if you are on stage gigging at several different venues each month, keep a notebook page for each place you play as to the best settings for each set of room acoustics that you encounter. Each venue will have a slightly different setup because of curtain locations and distances to nearby walls, etc., and having setup notes will allow you to fine tune your amp to the room much faster than trial and error each time you are there.

Funny thing about bass amps is even when set flat they still have some voicing. For example I can set one flat and plug my bass driver into the instrument input and then into the direct amp-in jack and get a different sound with each!
