Using a Fender Rumble 100 and would like to connect it to our mixer board, which has Phantom power turned on. Will this cause any damage to the amp? Also, how should the Ground Lift button be set?

Any help would be appreciated!

the line out in that amp comes directly from a winding on the output transformer. the DC voltage from phantom power will NOT damage the line out circuit in the amplifier. Phantom power puts equal value voltages on pins 2 and 3 of the XLR to operate equipment needing external power. DC fed back on to the two terminals of the transformer will do no damage.
