I cannot tell. I have never played a strat with a "D" shaped neck.
Left handed diversed guitars are hard to find. I managed to try a "V" shaped Gilmour, I own a 62' custom RI who has fat 'C' shape which is really good and an American Standard.

I would have to try out a few as I can't remember what they feel like Mike. :D

Hi Ben:
I don't know exactly what guitar you auditioned, but the neck presently on Dave's guitar, as reproduced by the Custom Shop, is the neck which was swapped from his 1983 57V cream Strat and installed in the Black Strat in 2005. The original neck has a date stamp of 11-2-83. It was built by Fender employee Jose Moreno. The '57V' designation is taken from that given to the new series of guitars ['57 vintage reissue] that Fender began to produce in 1983 when they rose out of the ashes. Dave acquired two guitars. One with a 'cream' finish and the candy apple red, which was to become closely associated with him. The neck is a C-profile the specs of which you'll find in the Custom Shop products section.

Ah yes, my bad :')
It had an American Standard Modern "C" shape. The guitar wasn't an exact replica of the Gilmour strat as it was black, even for the pu's covers. But it had the electronics that Dave has.

I have "D" necks on two of my Strats -- they were made by Fuji-Gen and used on the CIJ '65 Premium Re-issue guitars. They are absolutely the slimmest, fastest-fretting necks that I have ever owned (of any instrument brand). The profile is extremely shallow -- very thin and very comfortable. One neck is slightly "flamed" on the back and both have the nicest looking non-Brazilian rosewood fretboards I've ever seen. The vintage-spec fretwork is superb and if I ever see another for sale I'll happily grab it.

I did an experiment, by accident. The MIM tele I have has a slick poly neck, so I wax it with the rest of the guitar. A while ago, I forgot to polish the cloudy wax off the back of the neck, and used it.
Examining the neck afterward showed me where I put pressure on or rubbed the neck. I was surprised - it appears that most of the pressure is from my first finger base knuckle, not the thumb. And examining my hand, I even have a callus there. The only real rub spots on the back of the neck were from barre chords, where a D shaped neck probably would work well.
