Would it be possible to hook up a push-pull volume pot and a 3 way switch on a Strat as such? I saw some PRS guitars have pickup switching similar to this.

Push-Pull (Up)
Position 1. Neck Pickup
Position 2. Middle Pickup
Position 3. Bridge Pickup

Push-Pull (Down)
Position 4. Neck and Middle Pickups in Series
Position 5. Neck and Bridge Pickups in Series
Position 6. Middle and Bridge Pickups in Series

Everything is possible.

The 5-way selector offers more tonal flexibility than a 3-way.

The S1 switching system on the American Deluxe series gives you the following pickup combinations:

1. Bridge/Mid/Neck
2. Bridge/Mid
3. Mid/Neck
4. Bridge/Neck Out-Of-Phase/In Series w/ Tone Capacitor
5. Bridge/Mid In Series/Out-Of-Phase/In Parallel with Neck Pickup

He need 6 positions . 5 way switch is ...5 positions !

Plus some pickups in series .....He need a lots of contact and good chance to find the layout

My apologies but it's the only series/parallel push-pull wiring diagram for SSS I've had found on the Web.

I put a push-pull pot on the first tone pot (middle knob) of my Stratocaster. It turns on the bridge pickup, no matter where the pickup selector is set.

With this configuration, I can get all the normal sounds that a person gets with a 5-way switch, plus (wth the push-pull pot pulled up) neck + bridge and neck + middle + bridge. It's a great, (visually) discreet little modification.

The schematic is pretty close to what I want. I thought about using a 6-way toggle switch (Stew-Mac), just exploring other options at this point.

I have to say I love the concept of these. I have that on my Schecter J Bass (it's a common feature with most of their instruments) and I know Gibson has that option, too. I love the fact you can get a humbucker to sound like a single coil and vice versa. It gives you more options with one guitar.

There really is only 10 variations of pups selection with single coils...
If the OP is using stacked HB's then they'll be more possibilities..
But really, keeping the layout as simple as possible is best

OP could also use a superswitch instead of the 5 way...
An SS is basically 2 five way switches in one...

We would need a bit more info from the OP as to what music styles he is playing and as a live setting what are the parameters..
The guitar is .....? Model and pups ??

Rewiring is fine, but it can get not only complicated , but expensive.
Is there a budget parameter ??

The pickups will be Fender Custom Shop '69 Strat Pickups. I may just use the 6-way toggle switch in place of the standard 5-way blade:
