One thing to check out is whether/which pedals have a buffered/unbuffered bypass option. Try to make it unbuffered whereever you can.

A troubleshooting hint: Remove one random pedal. Does the problem persist? Remove another one. Repeat until problem is gone. Then you have identified one of the culprits. Now put all pedals back, but move that one pedal around to different positions. If that doesn't help, sell it to the gypsies too.

I now have a used Boss TU-2 tuner on the board. I currently have 10 pedals, but the One Spot daisy chain only accomodates 8. I plan on buying an additional daisy chain.
Is it better to hook this up to the current One Spot daisy chain of 8 or to the tuners power output?

Yep, I think you could, if the polarity is correct (find out first!!)

I always laugh at the prices some people will pay for a regulated power supply. It's a transformer, four diodes, a few capacitors and resistors and a voltage regulator. You can build one for about £15 tops. There's nothing "custom" about them, they all use the same VR's and the same design. There's not even any design work to do, the manufacturers of the VR IC's publish a recommended circuit diagram to use with each regulator device. The only difference is the amount of snake oil the marketing team rub into them. $180 for a PSU? Yeah, no thanks.
