Isn't that another name for permanent markers?
A friend of mine had his guitar signed by Satriani, with a permanent marker, and it does rub off.

You could spray a layer of Poly over the signature so it doesn't wear off.

No.

Not all "permanent" markers are created equal. For example, the Staedtler ones I was suggesting have an ink the solvent for which is methylated spirit/denatured alcohol, specifically so you can wipe them off slick surfaces such as cell and Perspex when you want to, but they remain otherwise fixed. For professional purposes. Google "Staedtler indelible pen" to find out more, or just look out for something like their Pigment Liner range, which will do what you want. For signing your guitar you want a nib size from the fat end of the range. This:

Other ranges and nib styles are available.

Of course, all pens that will write on a slick surface such as guitar lacquer do so by creating a film on the surface, not by staining it. And therefore they will all come off if you rub hard and long enough. Ultimately, the only sure-fire solution is to clearcoat over the top.

But a professional grade indelible ink will do the job well enough for most real-life situations. Choose the right ink.
