I have a back injury, so I stand until I start feeling it then use a guitar stool - so both.

Entirely dependent on the moment.
Practicing Classical requires sitting on a chair and elevating the leg with a foot stool, which is the position I use whenever I'm learning wether that be playing the Classical, Electric or fingerpicking an acoustic...
As much for comfort and proper posture when sitting for many hours.

Jamming with friends will be standing up since it's usually in a barn and motion is part of the experience....

This is another one that for me, it depends. If I'm just down in the studio trying to work out my chops, I'm usually sitting...I have an old office chair with one arm busted off that works great for such practice. If I'm on stage with the band however, I'm almost always standing. I say "almost" because there was a night a month or so back where I was just -beat-...really really bad day, so I spent most of the night sitting/leaning on the corner of the pool table that was behind me while I played. Actually about half way thru the second set, I actually pulled my mic stand over to the pool table so I wouldn't have to get up to sing...again, just a really bad day that night.

I get bunched up sitting in a chair, play best while standing.

I use the On Stage DT8500 Guitar/Keyboard throne when I need a compromise between sitting and standing.

I'm probably weird in that I have my strap adjusted so the guitar is the same height when I stand as when I sit. In fact I like to wear the strap when I sit. Thank goodness Mike Bloomfield made it okay to play with the guitar near your chin. :D
