Setting up a guitar I noticed that to get proper intonation on a couple of strings I had to set the saddle back almost as far as it would go. This made me wonder if there is more than one sweet spot along the travel of the saddle that will allow correct intonation.

Not that I know of, but I could be wrong. If I remember correctly the proper intonation point is exactly the halfway point between the nut and saddle at the 12th fret.

I set my intonation at the last frets.
I also set my amp for overdrive to tap harmonics vs the actual fretted notes.

I intonate with 12th fret harmonics. I think it works best that way.

Since most saddles have a beveled face on at least one side you can sometimes turn the saddle around to get the string-carrying notch a little farther from the nut. I had to do just that on a Les paul copy kit I built. Actually needed to do that on a couple strings but the intonation did come out right then.

There is only one sweet spot. String gauge has an impact on this sweet spot though.
The heavier the gauge the more the sweet spot moves towards the neck and vice versa.

when setting my intonation I usually fret the notes at the 12th fret and then check them against the 12th fret harmonics. In days long gone by I used to be able to do it by ear but I rely on my tuner a lot more now. I've only had a few times when I've needed to adjust the saddles, mostly when I've changed string gauges on the guitar HTH
