i dont see how it would. try it both ways and see what you like best.
i dont think there is a right or wrong way to do your pedals. i do think there is an undesirable way to do it in the tone results you'll get.

Toronado, from what I understand... You can easily run the MXR EQ out of the effects loop on that Peavey. But the OCD should be run straight out the front.

OD's, Distortions, and seething "death metal pedals" :wink: should NOT be run out of an effects loop - at least that's what I read on the internet.

Alright, that makes sense. So stuff like EQ, modulation, etc. are fine for running in the Effects Loop, while drive pedals are something you WANT the hit the preamp. Am I right?

Edit: I just read that for tube amps, some people like to throw in an extra boost in the Effects Loop to push the power amp tubes exclusively. My Peavey is SS so it's not anything I have to worry about.

Here's another question: Does a tuner turn off all other signals, including what's in the EFX loop? I'd like for the tuner to have the purest signal possible. I understand that ideally it's placed first in the chain.

i think there are tuners on the market that can do both, let the sig pass through and also block it off when in tuner mode. yes usually the tuner is first.
ive always just ran my effects straight into the front and go with it.

to jr - i think it would benafit the sound, ive experimented with some boost pedals but i dont think you would regain the punch of the valves... the boost pedal would probably work quite well... just play harder :P

tornado - most pedal based tuners have 2 out puts - one for true by pass where when the pedal is on the signal still passes through, and the other output cuts the signal...

Thanks rok, would using the signal-cutting output have a negligeable effect on the overall sound when the tuner is turned off?

Run anything you like into a effects loop. The only thing I don't recommend you do is put the speaker out of a amp into the effects return. Though I used to run a Marshall mini stack headphone amp from the headphone out into the effects return. To completely bypass the front end of the amp, substituting the headphone amp for the amp sound.
Sounds not rules.

Personally I don't like effects loops. I don't even own a amp that has one these days. I did, when I was younger, run a wah in the loop, as well as a distortion unit. It's ok you can still get usable sounds out of em, different but usable.

ive used the plannet waves pedal tuner and the whole range of boss pedal tuners, when all the pedals were off (boss tuners and plannet waves) there was no noticable difference but on the plannet waves pedal when in bypass and turned on i noticed a sligt fuzz on the sound, similar to the sound you get if you run speaker cables next to your signal cables...

i havnt tried other brands of tuners, so i cant really comment on all the tuners but most circuits are designed around the same principal so they should be compleatly off when off, if it sais true bypass on it then there will be no effect on the sound when its off...

just on the side, y some pedals seem to still change the sound even when they are off(ive noticed this with the boss pedals) is that some times in the non true bypass pedals the pre amp section remains active even if the pedal is off... in a tuner there is no pre amp so it shouldnt make much of a difference...
