ok i am wondering if i can play a epi valve jr. into the input of a line 6 spider 3 15 watt? or if it will blow up or whatever.

Yes. You never go from the output of one amp to the input of another.

The Valve Junior Hotrod has a dedicated preamp/reverb out socket.

As long as yours has that you're ok to go into another amp.

Don't go from the speaker out into another amp. Can't see it on the regular valve junior spec's. Is there a difference between the two amps?

Unless your amp has a "slave out" it isn't advisable to run one amp into another through the speaker output.If you want to run 2 amps together the best way to do it is to "daisy chain"them together,thats to run a patch cable from the input of one amp to the other.That only works if one amp has 2 or more inputs.
