Do you need to take the strings off of a guitar to use fretboard conditioner?

I wondered the same thing. I actually heard or read that you don't want to remove all the strings at once due to the fact that you would lose neck tension or something. However the next time I change strings I am going to remove all of them , and while they're off I will do a thorough job of cleaning my fretboard. I didn't realize how dirty it was (it must've been returnd as a starter pack several times) untill after I changed my strings so I had to do a half-#ssed job with a q-tip and Dr. Ducks axwax. It looks way better now that its dark brown/reddish.

I'd say it's advisable.

Most people do it when they're changing their strings anyway, two flies one belt.

I remove all the strings every time I change my strings. I have no problems with the necks on my guitars.

All my guitars have Rosewood fretboards and this is how I take care of them.

Using a piece of 0000 steel wool I lightly go over each fret going with the grain. Then with a q-tip I apply Gerlitz Guitar Honey. Let it soak in for a few minutes. Re-apply, and leave it sit again. Repeat for as many times as it takes for it to leave small pools. Take a paper towel and rub off all the excess, wait to see if any more comes to the surface. If it does wipe it off too.

Re-string, Tune, check your setup, and play!!!!

I condition my fretboard whenever I change the strings. I have always removed them all and so far have not had a problem. It is very easy to do once there is an open fretboard. I use a method almost the same as the one CAFeathers described.
