Post SRV, who was deservedly influential in the blues genre, the Strat has clearly won out as the instrument of choice for blues musicians. During the sixties, I've heard, guys who played blues tended to play telecasters, think of early Clapton or Muddy Waters. Personally, if you say blues, I think strat or National Tricone/single cone guitars. I think National, maybe more so than any other instrument, for me, oozes the blues. But that's just my opinion, of course.

You should be able to play the blues on just about any guitar. Hell, I've seen it played on a couple of wires nailed to a porch. Having said that, my preference is, for electric blues, a Strat, for acoustic, a nice resonator guitar works for me.

I have a piece of crap 3/4 size yamaha acoustic that sounds terrible. Except for blues. For blues it sounds better than any other acoustic guitar I've played. It's perfect. :)
