How do you get your own "signature" tone out of your amp? Personally I like switching my Strat to position 2 and turning off the treble and bass while knocking the middle up all the way. :D

Mine's mostly the other way. Middle half way, Treble and Bass full on, not always it can vary with acoustic circumstances. Whatever floats your boat man.

I have 4 settings which i mainly use, which 1 one use depends on what i'm playing.
My fave tho on my express is to use Crunch with gain on about 2 oclock
Bass on 11 oclock mid around 8 and treble about 11 also.
Dials arent numbered so have to use clock positions.
Then I use the footswitch to add in contour or reverb.
Unfortunately volume is down low cos even set 5w it can blow the house down :D :D

I use two Fender Stage Combos (a 100 and a 112se split from an MXR chorus pedal).

The 112's Gain at about 5 or 6, Treb 6, Bass 7, contour 4, Volume between, 2-3 Reverb 4. Clean side, Vol 3 teb 6, mid 8, bass 7, mid shift on.

The 100's Drive at about 7, Vol, 2-3, Treb 5, Mid 9, Bass 6, Reverb 3. Clean side Vol 3, Treb 6, mid 7, bass 7, Rev 4.

Then there's a TS 9, for a little tube-esqu drive. Then I mix and match clean and dirty channels depending on what song Im playin.

I have a few amps with just a volume, but the amps with an EQ:
All knobs point to the right at 8 (or 3 o'clock).

I'm looking for a warmer tone so my Strat's tone controls are about half-way and the pup switch is at 5 or 4 usually.

Cyber-Champ - Gain 5, Vol 5, Treb 8, Mid 5, Bass 4

Hmm. Looking at the settings it doesn't look like it should sound warm, but the amp emulation ('65 Princeton I think, it's a stored setting) not only warms it up but makes it a little muddy which is why the treble is so high.

For most rock applications

Strat switch in the bridge position (DiMarzio Tone Zone humbucker)
Volume and Tone controls on the guitar "dimed"

Bass on the amp set at 4
Mid on the amp set at 6
Treble on the amp set at 5

I use most of pickup combinations on my strat, but tend to favor the neck, especially for rhythm. My strat is an EC strat so I have the tbx knob on about six, guitar volume between eight & ten, the mid-boost goes up to three on the bridge pickup only (kind of a humbucker sound).
From there I go to a Dunlop Crybaby Classic (used seldom) and a MXR/CAE 402 boost/overdrive. The overdrive is used for a lead boost with the bridge pu only because w/the other pu's it turns to mud, the boost for a lead boost on all other pu's. Them comes the amp - Traynor YCV40WR that has the tubes changed out: 3 Electro-Harmonix 12AX7's and a matched pair of SED SV6L6GC power tubes (they cost about $20 more than most standard replacements but I would recommend them to anyone looking for a more vintage tone from their tube amp.)
I mostly use the drive channel on these settings:
Gain: 3
Volume: 5 (It gets pretty loud here!)
Treble: 5
Middle: 6
Bass: 7.5
Presence: 3
Reverb : 2 or off when recording.

It is a great blend of blues & psychadelic 60's rock! :D

Digitech Death Metal, equilized or a tiny bit up on high

Telecaster, well, it depends on what I want. Modern mainstream sound, tone all up, volume 2/3, on any position. Bright sound, tone up and volume up, bridge position. Muddy, volume up, tone down, neck position.

The list goes on. It truly is suprising how versitile the Tele is.
