I am looking at getting a loaded Custom Shop '54 Pickguard and would like to know if I can simply drop it into my Highway One HSS?

If so, how will the larger route from the humbucker effect the tone from new single coil bridge pickup.

As an aside, does anyone have the Ohm ratings for the factory Highway One pickups? I can find the Atomic Humbucker but not the single coils.

Yes, you can just change the pickguards. A lot of Strats with the SSS setup are actually routed for HSS. You should not notice any difference.

The man is right it should just drop right in. If you are worried about weird harmonics/microphonics from the extra space under the guard just put some cotton batting in there to fill the space.
