If I add a single coil sized Humbucker (Seymour Duncan, dual blade) into the bridge position of an otherwise stock pickup equipped Strat (Fender SCs in middle and neck positions), which pots should I use for best results?

Right now there's 3x 250k installed but humbuckers need 500k pots.
I've got volume and 2x tone control.

What disadvantage is to expect if I simply use 3x 250k???

Duncan designed the pickup you are considering around the premise that 250K pots will be used. There wouldn't be any disadvantage.

I also read a tip to try 300K pots in this circumstance but they might be more difficult to find.

300K linear pots are what Gibson has been using for a while now for tone pots. They will still strangle a full sized HB's frequencies quite close to that of a 250K pot. If a linear pot is used for volume, there will be 'bald' spots and ramp ups/downs when dialing the pot.

The larger value the pot, the more it will spread out a pickup's frequencies. In this case, it will slightly dip the midrange in favor of lows and highs.
