The biggest improvement is that it sounds less harsh...I hope there's not a really obvious reason to it that makes me look stupid. It's a Blues Junior III.

Would the amp sound less harsh to the audience as well?

Acoustically coupling the cabinet to the floor tends to improve the overall bass response as the floor acts like a funnel to keep the low frequencies more localized and focused.

Could also be that it is heading more directly at your ears on a stand, less so on the floor. Highs are very directional, lows less so.

Theoretically, you will get some cancellation of certain frequencies of the low end when the amp is on a stand (depends on the height of the stand). Some of the sound from the speaker bounces back up off the floor to mix with the direct sound from the speaker. (It also reflects from nearby walls but we're concerned with the floor here.) Some frequencies will be 180 out of phase when the 2 sources (direct and floor reflection) recombine and there is cancellation. For a speaker 2ft off the ground the centre frequency is approx 140hz. This is in the low end so an absence of low end leaves you with relatively more mids / tops which could be heard as less warm / more harsh. The cancellation frequencies go up as you get nearer the floor. I'm not saying this is the only explanation but it may be a contributing factor.

It's worthwhile to get a long guitar cable and walk out to where members of the audience will be standing because the problem is that while it might sound OK to you now that it's eq'd to sound good while shooting into the back of your kneecaps, out in the audience it might be like the proverbial ice-pick and taking people's heads off.
