I'm trying to figure out if I'm just driving the speaker hard at the wrong frequency, but my new '65 Re-issue will make a distinct buzzing if I play a low E open. Granted, this isn't until I turn the volume up past 5, but is this normal?

Check and make sure that all the screws, nuts and bolts are tight. Don't overtighten the speaker bolts. Make sure that there are no wires vibrating against anything inside the amp. You could also have a resonance in the room. Try playing the amp in a different room and see if it does the same thing. Cabinet/room vibrations can be tricky to troubleshoot. Good luck.

+1 on the above.

After checking inside the cab for the above move the amp around to see if it is room resonance. This happened to me.

You will also find several threads where some of us had a lot of trouble with the 5AR4 rectifier tubes "Buzzing"

Bet you give yours a light shake and it rattles.

I used the JJ in one and a Sovtek in another DRRI and it solved the problem both times.

I know of at least two other posters here who had the very same trouble and it was the rectifier tube.

I have the same isuue..
I here you on the acoustics of the room.. & yes things buzz in the room from sound waves reverberating.. But the sound i am referring to is coming directly out of the amp & speaker.
& OD Petals aggravate it..

Bill,
This is my first "real" tube amp...I just re-kindled my guitar playing interest from about 30 years ago. I left off playing with a Gibson LS-6 & a pevey studio pro.. around 1980-81. I was 11.

What is the best way to trouble shoot a bad tube?.. I was told take a pen & tap on the tube & check if the sound transmitted through to the speaker..- dont know if its BS?

You can GENTLY tap the tube with the eraser end of a pencil or a wooden chopstick. Sometimes the tube is so microphonic, you can tap the chassis right next to the offending tube and hear the "thud, thud" through the speakers.

By rotating the tubes around, you can often time get a noisy tube out of a sensitive position (like the input stage or V1) and reduce the overall effect of the loosy-goosey tube. Prolly, the best way is to rotate a known, good tube into each corresponding position and type--- and see if this reduces the bad noises.

BTW... this works ok for smaller gain stage tubes, like 12AX7 or 12AT7. But, I wouldn't tap rectifier tubes or power output tubes too much. Best to just try substituting known, good tubes into these positions.

Just to be sure... this isn't a fret buzz issue, right?

In my case, I'm pretty sure it was the speaker or the cabinet. I turned it off, unplugged the speaker, and plugged it into my Hot Rod Deluxe, and it still buzzed. The buzz was definitely coming from the speaker and not parts of the chassis or tubes. I didn't feel like messing around with a new amp that's not working right, so Guitar Center took it back after they tried it out and concurred that a new amp should not be buzzing like this one.

In my case it’s not the frets or a loose speaker buzzing. & It is mainly caused when I use a (non-compatible) OD petal in the vibrato/reverb channel.
The problem is discussed in detail here:

Some people mod the amp to make it more accepting…. but I am not into it.

I found that the Fulltone Fulldriver-2 works well. You can dial in any sound with it, to avoid a crappy buzzing sound. A diverse petal. (The Boss BD-2 & the OS-2 were horrible)

I too replaced a Boss BD-2 with a Fulltone FD-2M. Even after modding it, the BD-2 was too fizzy.
