So. I replaced the tubes on my blues jr today and I was wondering which of the preamp tube locations controls the lows, mids, and highs, respectively. Because I want to boost the lows in the amp, so I got a higher gain preamp tube instead of getting them all the same. So does anyone know which one a I should put it in the boost the lows? Thanks.

Preamp tubes amplify all frequencies more or less equally. There is no specific tube for bass, treble, and middle. Bass, treble, and mids are adjusted by twirling the tone control knobs to your liking. As far as where to put the high gain tube, you only have two options, V1 or V2. Try it in both and see which sounds best to you. Don't put it in V3.

Just to said that long plate valves promote bass but more risk of microphonic. Usually short plate are prefered.

While a long plate tube may have better bass response, I doubt that it is enough to make a significant difference in bass. That is still what the tone stack is for. But the point is, no one tube is strictly responsible for bass, middle, and treble which the OP needs to understand.
