I have a Blues Deluxe Reissue.
How do you know when is time to replace the tubes?
I really would have looked it up, but I cannot find a search box in the forum.

P.S. Does amp hissing have anything to do with it?

It depends on use and if you are having issues! Are you having any issues and what are they?

I use it five times per week for 1.5 hours.
Have had it for almost five years.

Yes, it is most likely time for new tubes! Five years is good! Change them before they go bad and burn out something else! Get the amp biased!

There should be a search box in the upper right of any page on this forum (Fender on upper left, search box far right). It even says "Search Fender.com" inside the box, with a magnifying glass beside it. This topic has been discussed many times here, you should find a lot of posts using the search.

As far as when to change tubes, if you notice a loss of clarity, or the sound has degraded from your amp, it might be time to change. Is the "hiss" any worse than it has always been? Most tube amps have some hiss, that's normal. What tube(s) do you think are microphonic, and why?

There is no set time to change tubes, and depends on whether or not they have been biased hot and/or run hard (amp wide open all of the time). My HRDlx has the original tubes and is over ten years old, still sounds great. The bias in the HRDlx was perfect from the factory, and I used to play it often, with pedals and very loud. Also, look at your tubes and see if they have burnt spots inside on the glass, or if they look OK. You can get a lot of years out of tubes if they are setup right. If you do find the need for new tubes, like Cap47 said, have the bias set correctly. :)

Edit: P.S. Using your amp 1.5 hrs/day everyday for five years is only 2737.5 hours on the tubes. I think that +/- 5000 hours would be a reasonable number of hours to expect from properly setup tubes.
