It's poular opinion that distortion and overdrive pedals are usually best placed in front of the amp rather than in the loop. It's also popular opinion that delays, chorus, flange, etc. can benefit from being in the FX loop. I suggest you try different configurations and see if you can hear a difference and then choose whatever sounds good to your own ears.

Just think of how awesome a guitar player you could have been by now if you had only spent the last 10 years practicing instead of obsessing over pickups and roasted maple necks.

+1
That's how I and millions of others have our pedals arranged.

In my case it's: Wah, overdrive, distortion in front of the amp. Chorus, flanger, and delay in the loop.

Delay pedals really seem to benefit the most from going in the loop if you want clear sounding repeats. A delay in front of an overdriven tube amp will have its repeats sounding like mush. Some like that sound though, so there are no rules.

Since most amp distortion is generated in the preamp section of an amp (especailly amps with switchable clean and distortion channels), it all boils down to if you want your effects before or after amp generated distortion. Hence the two places they can be placed: at the front (before distortion) or in the loop (after distortion). Generally speaking, effects that manipulate gain and volume like overdrive, distortion pedals tend to sound best in the front, while time based and modulation pedals like delay, chorus, flanger sound best in the loop. For example when deciding where to put your delay pedal, ask yourself "Do I want to distort my delay's repeats, or do I want to repeat my distortion?"

Here are some helpful links that can describe it in more detail than I can:

Hi Guys,
You know I had never tried the "loop" on my Hot Rod Deluxe... You know what, It makes a difference! All of my "modulators" chorus, flanger, Delay etc sound much cleaner. So for me it really did make a difference. YMMV

When I use my POD, I think it sounds better plugged directly into the "Power Amp In" on all my amps with that input, except for the Deville. The Deville sounds better with the POD plugged into the input jack. The Deville has higher gain preamp tubes than my other amps, that might be why. The POD has a volume control, which I think is a must if you plug straight into the "Power Amp In" jack.
