I don't know much about it either, but this guy does:

There's some really good reading on that page.

I'm not an amp tech, nor do I know much, but I have learned how to bias my Blues Deluxe RI.

Biasing sets the environment in which your amp operates. A low or cold bias will cause your amp to sound flat. A hot bias can cause it to sound crisp or to breakup sooner. If you bias too hot you can destroy the tubes. Too cold will cause tone problems, but I forgot what sort. Also, biasing hot will cause your tubes more stress and lead to you needing to replace them more often. Obviously, biasing directly affects your tone and is one way of altering the sound of your amp. Changing the tubes without biasing them can destroy the tubes or leave your sound cold or with too much bite.

There are instructions for biasing a Hot Rod/Blues Deluxe series amp at http://studentweb.eku.edu/justin_holton/ This is where I learned how to bias my amp. As is stated at that website, there is some danger involved with biasing your amp, if you don't follow the proper procedures and take the proper precautions.

Having said all of this, I would say that learning how to bias my amp has been fantastic as I now feel like I can greatly alter the sound quality of the thing. I don't have to rely on the ear of someone else.
