Hi everyone, and thanks in advance. I am using a Boss ME-80 multieffects unit with this amp, and a Strat ultra with lace sensor pickups. Any suggestions for settings on clean and dirty channels to obtain the result I want?

The Strat Ultra with Lace is a great guitar. One of the best I ever try. Do not sell it, never.

With a better amp and better effect, you'll discover how great is your guitar tone.

About Marshall sustain and distortion, you must use a Marshall amp .....

I use my Boss ME-70 for practicing with headphones; its a great way to really learn whats possible with the ME.
I have found when plugging the ME into the amps input, some of the characteristics get cut out by the amps capabilities.
Be careful with the pre-loaded pre-amp settings, they go from 0-12 in a heartbeat when they kick in. It takes a bit of time to dial in the settings that really work w/ any amp.
I like using compression for sustain, boost or classic for OD and analog delay or reverb to taste.
I use the pre-amp sparingly for gain and mid freq balancing.
The difference between our model ME's, is I think yours is digital adjustments and mine has the traditional style pedal knobs.

That guitar should sound great plugged straight into the amp on 10.

In my experience almost any crunchy amp can give you sustain ... if you crank it up.
There is a definite correlation between volume and sustain and controllable feedback that can be obtained other ways ... sort of .... to a limited extent .... but if you want the kind of sustain and feedback where harmonics blossom into a glorious sounding Rock-God squeal you're going to need to dime it.
You can encourage such things using compression, chorus, drive, etc. but for the most part you're not going to get the real deal at bedroom volumes.
It just doesn't happen.
People will tell you otherwise but if they actually had any real first hand experience with a cranked Marshall they'd be forced to admit it's just wishful thinking to believe you can get that sound any other way.

As for the actual Marshall style tone, you'll want an EL34 amp with a preamp section that caters to that sound.

There used to be a pedal that was supposed to simulate a cranked Marshall stack but they're out of production. The pedal was called the Guv'nor. You can still get clones of that pedal but in my experience the Guv'nor was just another distortion pedal. I have one of those clones. They all sound the pretty much same to me. Different flavours of the same thing.

I do not believe the essence of Marshall magic lies in the grind/drive but rather in the amp itself when dimed.

This pedal does a nice job of sounding like several Marshall models, and also has a separate "Tube Screamer-esque" circuit (pre-gain button). So it's like 2 pedals in one. Expensive, but sounds great:

THANKS!!! You've all been so helpful. I will never sell my strat ultra with lace sensors. I've played many different guitars of all brands, and nothing touches it. I've had it since about 1995-1998.

+1 (but with qualifications)

As mentioned, there are some multi-effects pedals available that include amp simulators with varying degrees of authenticity. I personally use the Zoom G-1X but devices from Boss, Digitech, Wampler, and others are worthy candidates. There are two tricks to using pedals such as these: 1) you need to spend some time with the owner's manual and learn to manipulate the presets to dial in the exact Marshall vibe you seek, and 2) when using these pedals it's important to set the amp's controls for a straight clean tone and flat frequency response. Let the device do the "heavy lifting" and use your Champion as a linear power amp. Use the reverb if desired but leave any boost/fat/OD switches off.

Ever heard of the Matchless Hotbox? I have version one. Demo is of version 3. Runs off two 12AX7. I like the 7025 or 12AX7A has they are designed to be quieter (heater hum-wise) than the plain 12AX7.

It has a nice varied level of crunch. Gain OD. More selective and sensitive than most solid-state pedals. Except, maybe the original Tube Screamer with the NEC 4558 chip.

About 20 years ago, a friend and I DIY a Hotbox, but using the very linear Tungsol 5687W. That pedal had much more linear response to the controls. And a midrange tone to die for. I'm still thinking about redoing my Hotbox to take 5687 tubes.

You could start with the #8 selection in the ME-80's Preamp section. That's called "Stack", which is a 100-watt Marshall head tone. Patch # 3-2, "70's Hard Rock", is another place to start. Read the manual, start with the factory patch, then save it to the USER area after you tweak it to taste. Most of the factory presets tend to sound better when you back off from the usual maximum settings they give you. There's often too much gain, too much reverb, too much of everything. This can demonstrate the effects well, but makes your guitar sound like a beer commercial. Not that there's anything wrong with that.
