I play roots rock,blues & sometimes a little jazz. I have a hand built deluxe I've been playing for years now(great amp) I am looking for input. In this type of amp what tube and speaker for that matter do folks feel should help me achieve a true old school tweed deluxe tone?

Tubes are pretty subjective (I favor old-school NOS) but for true vintage speaker vibe, I don't think you can beat a Weber 12A125. If you can find an original (ie: made in Chicago non-reissue) Jensen P12Q, that would be even better. YMMV

I understand taste in tubes will be personal. This amp has had JJ's in it. 1-12ay7 1-12ax7 2-6v6 and a gz34. That is what the builder suggested.it has murcury transformers and a Jensen c12n. I changed the hi output 6v6 tubes the maker originally put in for me with the JJ's. I use this amp in Phoenix Az area for club dates and do not want it to break up at to low a volume. I did replace the gz34 with a old 5y3 (not sure of the brand off hand) but it seems like the straight (no booster/dirt box) is MUCH. Cooler when I dig in to a cord. Now I wonder what other changes might make things sound even cooler in a hairy vintage way?
